PT-141

These days, listen to any health podcast and you’re likely to hear discussions about peptides and how they can help you lose weight and build muscle with ninja precision. But one benefit often seems overlooked: they can boost libido in both men and women, as well as improve erectile function in men. While many peptides claim to improve your sex life, one prescription peptide—PT-141—is a real star.
”PT-141 is a revolutionary treatment for enhancing sexual arousal and libido in both men and women,” said urologist Dr. David Schusterman.
Unlike drugs that claim to have an aphrodisiac effect or promote blood flow to the genitals (such as sildenafil), the synthetic hormone PT-141 acts directly on the source of libido: the brain. This peptide stimulates specific receptors in the brain that are naturally activated during sexual arousal. “Unlike other erectile dysfunction drugs that act on the vascular system, PT-141 acts directly on the central nervous system,” added Shusterman. “This is a completely new solution.”
Jim Staheli is an orthopedic surgeon and medical director at Broad Health. He is an expert in men’s health, including issues such as decreased libido and functional medicine.
PT-141, also known as bremelanotide, is a peptide drug that binds to melanocortin receptors and may enhance libido, improve sexual function, and enhance sexual satisfaction. Dr. Jim Staheli (1, 2), medical director of Broad Health, notes that the most common route of administration for PT-141 is by subcutaneous injection into the abdomen or thigh. Some pharmacies also offer nasal sprays or tablets that can be used in combination with other sexual health-enhancing medications (such as oxytocin and Cialis).
This peptide may offer multiple benefits for sexual function. Shusterman suggests that this non-hormonal therapy could be effective in improving the condition of people suffering from both sexual dysfunction and decreased libido.
”PT-141 can activate the MC4R and MC1R receptors in the central nervous system, which play a vital role in controlling sexual arousal and libido,” said Dr. Alia Moore, a board-certified sex therapist.
Without motivation and desire, it’s difficult to experience sexual arousal. PT-141 can increase levels of three neurotransmitters: dopamine, norepinephrine, and oxytocin, which are essential for libido, motivation, and sexual arousal. Dopamine stimulates libido, while norepinephrine and oxytocin promote sexual arousal (3). PT-141 can also increase norepinephrine levels, thereby improving mood (4).
“Unlike traditional sexual dysfunction treatments, which primarily focus on improving blood flow or hormone levels,” Moore said, “PT-141 addresses the underlying neurological factors that cause low libido, providing a targeted and potentially effective solution for increasing libido.”
This is true for both men and women with chronic hypoactive sexual desire disorder. In 2019, the FDA approved PT-141 for the treatment of hypoactive sexual desire disorder in premenopausal women to improve libido and sexual arousal (3). The drug is also currently used in men as an off-label treatment.
Moreover, any type of sexual dysfunction can cause discomfort. Seeking treatment can help improve mood and well-being, thereby further enhancing sexual health (5).
Recent studies have shown that PT-141 may help treat erectile dysfunction in healthy men (6, 7). Early studies in rats have shown that its mechanism of action involves activating melanocortin 4 receptors (MC4Rs), which in turn stimulate the nerves in the corpus cavernosum of the penis, thereby helping men achieve an erection (8).
Stachley argues that stimulation of melanocortin receptors causes increased arousal, which facilitates the physiological process of erection. “This leads to selective transmission of nerve signals to the penile tissue, which stimulates the release of nitric oxide, promoting relaxation of penile smooth muscle, vasodilation, and swelling of the genital tissue.”
The role of the central nervous system may offer additional benefits. Moore adds that, in particular, it helps manage psychological factors that can contribute to erectile dysfunction (ED), such as stress or anxiety.
”This neurological approach makes PT-141 a potentially effective option for patients who do not respond well to traditional erectile dysfunction medications or who also suffer from erectile dysfunction and other sexual disorders, such as decreased libido,” Moore said.
Any medication has the potential to cause side effects, but that doesn’t mean you’ll definitely experience them. The most common side effects of PT-141 include (9):
Stahelli stated that people with heart disease or uncontrolled high blood pressure should avoid PT-141, as it can increase blood pressure. He also warned that people with severe liver disease or kidney failure should also avoid its use, as these conditions can slow the elimination of PT-141 from the body.
Staheli warns that PT-141 should not be administered more than twice a week and no more than eight times a month. Excessive use may cause skin hyperpigmentation. If you have high blood pressure, PT-141 should not be used concurrently with PDE5 inhibitors (such as Viagra and Cialis), as this may further increase blood pressure.
The onset of PT-141′s effects depends on the route of administration and typically occurs within 30 minutes to 2 hours. “The onset of PT-141′s effects varies from person to person depending on factors such as dosage, frequency of use, and health status,” said Shusterman. “For some people, increased libido and sexual arousal occur very quickly after taking PT-141, while for others, it may take longer to feel noticeable changes.”
PT-141 is a prescription medication that can increase libido in both men and women. It may be a good option for those who have unsuccessfully tried other traditional treatments for low libido. Please consult with your doctor to determine if this peptide is right for you.
